A simple procedure for the purification of active fractions in aqueous extracts of plants with allelopathic properties Acta Botanica
Borghetti,Fabian; Lima,Elisa Coutinho de; Silva,Lucas de Carvalho Ramos.
Most studies conducted to test the allelopathic activity of plant parts have made use of water as solvent. However, the presence of polar, water-soluble substances, such as proteins and carbohydrates, tends to hamper the purification of active compounds. In this study, we present a simple purification procedure that separates the active fraction of the extract from the undesirable substances, thus facilitating the search for active molecules through standard chromatographic methods. Aqueous leaf extracts of three Cerrado species (Caryocar brasiliense, Qualea parviflora and Eugenia dysenterica) were prepared at 5% concentration (w/v) and stored at 4ºC (crude extracts). After 24 h, these solutions were filtered and freeze-dried. The powder obtained was...

Effects of extraction method and storage of dry tissue on marine lipids and fatty acids ArchiMer
Sardenne, Fany; Bodin, Nathalie; Metral, Luisa; Crottier, Anais; Le Grand, Fabienne; Bideau, Antoine; Brisset, Blandine; Bourjea, Jerome; Saraux, Claire; Bonhommeau, Sylvain; Kerzerho, Vincent; Bernard, Serge; Rouyer, Tristan.
Various protocols are currently used to study marine lipids, but there is a growing interest in working on dry samples that are easier to transport. However, reference protocols are still lacking for dry samples. In order to make recommendations on this use, lipid classes and fatty acids (FA) obtained from six analytical protocols using two different tissue states (dry vs wet) and three extraction methods (automat vs manual potter vs leaving the solvent to work on tissue) were compared. Three dry storage modes of tissue (freezer vs gas nitrogen vs dry room) during one and three months were also compared. These comparisons were made on seven marine species with different lipid profiles, including fishes, crustaceans and mollusks. Lipid classes and FA...

Enhancing the aqueous solubility and dissolution of olanzapine using freeze-drying BJPS
Dixit,Mudit; Kini,Ashwini Gopalkrishna; Kulkarni,Parthasarthi Keshavarao.
The aim of the present study was to develop an olanzapine freeze-dried tablet (FDT). The solubility and dissolution rate of poorly water-soluble olanzapine was improved by preparing a freeze-dried tablet of olanzapine using the freeze-drying technique . The FDT was prepared by dispersing the drug in an aqueous solution of highly water-soluble carrier materials consisting of gelatin, glycine, and sorbitol. The mixture was poured in to the pockets of blister packs and then was subjected to freezing and lyophilisation. The FDT was characterised by DSC, XRD and SEM and was evaluated for saturation solubility and dissolution. The samples were stored in a stability chamber to investigate their physical stability. Results obtained by DSC and X-ray were analysed...

Formulation and in vitro evaluation of taste-masked oro-dispersible dosage form of diltiazem hydrochloride BJPS
Jagdale,Swati Changdeo; Gawali,Vaibhav Uttam; Kuchekar,Bhanudas Shankar; Chabukswar,Aniruddha Rajaram.
Diltiazem hydrochloride is a calcium channel blocker generally indicated for the treatment of angina and hypertension, and it is extensively metabolized due to the hepatic metabolism. Formulation of diltiazem hydrochloride into an oro-dispersible dosage form can provide fast relief with higher bioavailability. The bitter taste of the drug should be masked to formulate it in a palatable form. In the present work, an attempt was made to mask the taste by complexation technique, with a formulation into an oro-dispersible dosage form, using superdisintegrants Doshion P544, crospovidone (CP) and sodium starch glycolate (SSG). The complexes of diltiazem hydrochloride with β-CD (1:1 molar ratio) were prepared by kneading, co-evaporation, co-grounding,...

Freeze-dried Spermatozoa and Freeze-dried Sperm Injection into Oocytes OAK
SUZUKI, Hiroshi; 鈴木, 宏志.
Attempts to freeze-dry spermatozoa are not new. Successful conception and full-term development using freeze-dried spermatozoa were reported in the cow and rabbit prior to 1960. However, these preliminary results have not been confirmed. Spermatozoa become defective in motility after freeze-drying and are unable to fertilize eggs both in vivo and in vitro. However, in 1998, freeze-drying of mammalian spermatozoa was demonstrated without loss of genetic or reproductive potential. Freeze-dried spermatozoa support normal development when injected directly into oocytes by means of an intracytoplasmic sperm injection (ICSI) technique. Recent investigations of freeze-dried sperm have focused on the factors affecting freeze-drying in spermatozoa in an effort to...

Freeze-drying of ampicillin solid lipid nanoparticles using mannitol as cryoprotectant BJPS
Alihosseini,Faezeh; Ghaffari,Solmaz; Dabirsiaghi,Ali Reza; Haghighat,Setareh.
abstract Solid lipid nanoparticles (SLNs) are interesting colloidal drug-delivery systems, since they have all the advantages of the lipid and polymeric nanoparticles. Freeze-drying is a widely used process for improving the stability of SLNs. Cryoprotectants have been used to decrease SLN aggregations during freeze-drying. In this study Ampicillin was chosen to be loaded in a cholesterol carrier with nano size range. To support the stability of SLNs, freeze-drying was done using mannitol. Particle size, drug release profile and antibacterial effects were studied after freeze-drying in comparison with primary SLNs. Preparations with 5% mannitol showed the least particle size enlargement. The average particle size was 150 and 187 nm before and after...

NaCl stress impact on the key enzymes in glycolysis from Lactobacillus bulgaricus during freeze-drying BJM
Li,Chun; Sun,Jinwei; Qi,Xiaoxi; Liu,Libo.
Abstract The viability of Lactobacillus bulgaricus in freeze-drying is of significant commercial interest to dairy industries. In the study, L.bulgaricus demonstrated a significantly improved (p < 0.05) survival rate during freeze-drying when subjected to a pre-stressed period under the conditions of 2% (w/v) NaCl for 2 h in the late growth phase. The main energy source for the life activity of lactic acid bacteria is related to the glycolytic pathway. To investigate the phenomenon of this stress-related viability improvement in L. bulgaricus, the activities and corresponding genes of key enzymes in glycolysis during 2% NaCl stress were studied. NaCl stress significantly enhanced (p < 0.05) glucose utilization. The activities of glycolytic enzymes...

The influence of freezing rates on bovine pericardium tissue Freeze-drying BABT
Borgognoni,Camila Figueiredo; Tattini Junior,Virgilio; Ayrosa,Ana Maria Irene Bartolomeu; Polakiewicz,Bronislaw; Leirner,Adolfo Alberto; Maizato,Marina Junko Shiotsu; Higa,Olga Zazuco; Beppu,Marisa Masumi; Pitombo,Ronaldo Nogueira de Moraes.
The bovine pericardium has been used as biomaterial in developing bioprostheses. Freeze-drying is a drying process that could be used for heart valve's preservation. The maintenance of the characteristics of the biomaterial is important for a good heart valve performance. This paper describes the initial step in the development of a bovine pericardium tissue freeze-drying to be used in heart valves. Freeze-drying involves three steps: freezing, primary drying and secondary drying. The freezing step influences the ice crystal size and, consequently, the primary and secondary drying stages. The aim of this work was to investigate the influence of freezing rates on the bovine pericardium tissue freeze-drying parameters. The glass transition temperature and...
